Rothbury first appeared on a census roll in 1970 with a population of 394. By 1980, it had increased to 522, its peak population. The village lost more than twenty percent of its population between 1980 and 1990. Although it has increased slightly each decade since, its population at the time of the 2020 census was 462.

Nelson Green is acknowledged as the village's first permanent settler when he came in 1865. That year, he gave the Chicago & Northwestern Railroad (which later became the Pere Marquette and the Chesapeake & Ohio Railway) a right-of-way across his land. A railroad station was established as Greenwood Station. However, when a post office was established on January 19, 1876, with Clark S. Parks as postmaster, the post office was called Malta because there was another Greenwood post office in Michigan. The post office was closed on August 27, 1877, but restored as Rothbury on September 22, 1879, at which time Charles D. Arnold became the postmaster. When Rothbury was incorporated as a village, it took the same name.
